Cameron said there was no good evidence at this stage it was an illegal "secret mine."
"This is a national treasure, if you ask me," he said.
However, the Department of Foreign Affairs and Trade is examining potential "disruptive effects" on uranium resources in Australia if the project moves forward.
It's also raising questions about whether the government is being honest about any potential impacts, given the "lack of detail" it has offered.
"The industry certainly feels that the information is being provided in the most transparent way."
Cameron said government funding for this mining project is "much needed," but said funding was still needed to "fund a thorough environmental assessment."
Australia's Energy Minister Josh Frydenberg told ABC TV that he had been assured by government officials this information would be provided, but was yet to receive it.
Cameron on whether uranium mine ban would affect Australians
"I have to reiterate my concerns that as long as this particular mine exists, it is going to affect the future and that's why we have a clear regulatory framework," he said.
"This is a significant area where we know there's a lot of potential and I'm also worried about the potential for negative consequences for this region that's about 25 kilometres away."
But Cameron has said Australia would have to withdraw its ambassador from Cambodia.
Kenny MacInnes, former mining minister, warned the uranium mines could affect "the environment, the biodiversity and the culture of this region, especially due to the fact that we have a close cultural connection to it and so there's a direct impact on the people."
He said the project "can affect tourism, which we all need to continue to increase, because it is something we want to make sure we create good jobs."
Foreign Affairs Minister Julie Bishop said the government "was aware" about concerns with the uranium project.
The Energy Minister said the mine's location in Cambodia was the focus of the country's efforts to block uranium mining.
Cameron said the country was working "with our allies, with our partners to push back hard" against the mining project and work with foreign governments to work together on a "sustainable plan of action."
He also said Cambodia should work with the United States, Britain and other countries to stop further uranium mining, a point made by the United Nations.
Cambodia plans a report on the uranium project next month.
The Government is also reviewing its mining permit.
